It promises to be a big game for the Whites as they look to capitalise on an out-of-form Spurs side who were beaten in Gent on Thursday night.
The west Londoners reached the fifth round of the cup by beating Cardiff in round three before thrashing Premier League side Hull in the last round.
Spurs have found progress slightly more difficult as they saw off Aston Villa in the third round but had to come back from 2-0 and 3-2 down to beat League Two side Wycombe Wanderers in round four.
It is the first time the two sides have met since Easter Saturday in 2014, when Tottenham won 3-1 at White Hart Lane.
Paulinho's opener was cancelled out by Steve Sidwell before Harry Kane and Younes Kaboul scored in the second half to give the Lilywhites victory.

The Belgian right-back and his mates face the stiffest FA Cup challenge so far, although the last time the two sides met in the famous old trophy, Whites thumped the north London variety 4-0 in 2011. Odoi will also run into another set of pals: those from the national Belgium set-up he’s partnered in the past.
